Polyene macrolide antibiotic nanoemulsion: a proposal for the treatment of cutaneous leishmaniasis

نویسندگان

چکیده

Leishmaniasis is a neglected tropical disease that requires timely and inexpensive treatment. For this purpose, nanoemulsion with polyene macrolide antibiotic, or amphotericin B (NE-AmB), was developed. This study quantified the amount of drug permeated retained in intact lacerated human skin, simulating cutaneous leishmaniasis (CL) processes. Toxicity macrophage keratinocyte cell lines, activity against promastigotes amastigotes Leishmania tropica, vivo irritant activity, histological evidence evaluated. Results. The damaged skin 750.18 ± 5.43 567.97 8.64 µg/g/cm2, respectively. There no permeation. No apparent toxic effect observed HaCaT lines. IC50 NE-AmB found for 0.26 0.09 0.37 0.05 µg/mL, NE without AmB did show antiparasitic activity. formulation showed lower values on both parasite stages than solution. irritation, histology improvement We suggest may be candidate studies CL patients. Keywords. Leishmaniasis, Amphotericin B, ex permeation studies, vitro cytotoxicity, leishmanicidal Draize test, histology.

Oral Zinc Sulfate in the treatment of acute cutaneous Leishmaniasis

Background: Different local and systemic modalities are suggested in the treatment of cutaneous leishmaniasis (CL), but the pentavalent antimony compounds are still considered as the first line of treatment. Regarding to increase in clinical drug resistance, adverse effects and low compliance of patient, efforts for finding a more effective and safer drug continue. Objectives: To evaluate...

Topical treatment for cutaneous leishmaniasis.

Parenteral pentavalent antimonials remain the standard therapy for cutaneous leishmaniasis. More effective and patient-compliant topical treatments are an important alternative treatment for the localized self-limiting forms of this disease. Two paromomycin ointments are commercially available but their use is limited by either toxicity or lack of efficacy. Other topical formulations have been ...

Cutaneous leishmaniasis treatment.

The causative species of cutaneous leishmaniasis determines the clinical features and courses, and treatments. Intralesional or systemic antimonials are the gold standard for the treatment of these diseases. However, as for visceral leishmaniasis, other therapeutic options appear promising. Paromomycin ointments are effective in Leishmania major, L. tropica, L. mexicana, and L. panamensis lesio...

Trichomycin B, a polyene macrolide from Streptomyces.

Two polyene macrolide, trichomycins A and B, were compared by physico-chemical and microbiological methods. The two antibiotics were found to have the same molecular formula, C58H84N2O18 (MW 1,096), by elemental analysis and FAB-MS. However, 1H and 13C NMR spectrometry studies indicated that the hydroxyl at C-5 of trichomycin A located on C-9 in trichomycin B. Trichomycin B possessed lower acti...
